Detection, partial purification and characterization of Bacteriocin from Enterococcus species indigenous to Azad Jammu and Kashmir

Abstract
Study was carried out to isolate, partially purify and characterization of bacteriocin from enterococcus species indigenous to Azad Jammu and Kashmir. Isolation of bacteriocin from raw milk of buffalo was carried out by Spread Plate method and identified by biochemical test. Out of total milk samples (N=20) only samples (N=4) were found to be bacteriocin producing. The demonstration of selected strains was confirmed by 16S rRNA sequence of gene. The selected strains identified as Enterococcus faecium CGMCC 1.2136T, Macrococcus caseolyticus ATCC 13548T, Streptococcus gallolyticus sub species macedonicus ACA-DC 206T and Enterococcus faecium CGMCC 1.2136T. To partially purify the bacteriocin obtained from Enterococcus species. Ammonium sulphate precipitation method was used after the partial purification of bacteriocin to show its activity as per related enterococal strais and against identified microorganisms of hospital. High temperature stability (up to 100°C) was observed. Bacteriocin activity was also seen on larger range of pH (3 to 9). Treatment with proteolytic enzyme proved the protenecious nature, as the inhibitory property of bacteriocin was completely lost after being exposed to enzyme proteinase K.
