Rapid massclonal regeneration of chickpea (Cicer arietinum L.) through callus induction

Abstract
The plant tissue culture techniques have been used widely to multiply number of plant varieties, many successful propagations of plants in short time depends upon most efficient protocols in this regard there has been enough publications worldwide on micropropagation of chickpea, but limited word so far is done in Pakistan specially in Sindh province. To optimize the successful protocol the present study was planned and hypothesized to enhance the organogenic plant regeneration and rapid clonal proliferation of chickpea from callus. To stimulate callus initiation, the various treatments of 2, 4-D were used with amalgams of B.A.P showed significant results the max callus weight (1.14g) was recorded on C-IV medium. Amended various strength of B.A.P + I.A.A were used for shoot regeneration the maximum survival rate (91.0%) and shoot length (6.35 cm) was measured on SM-V, the minimum days to initiate (13.0) shoots were recorded on SM-IV medium however, the most number (5.0) of shoots were also recorded under the same concentration. The study found that SM-III had the most leaves (23). IAA had a significant effect on root initiation, with RM-IV showing the shortest time to initiate roots and RM-V producing the longest roots (5.30 cm) and the most roots.
